BioNTech Says Gotistobart Nearly Doubles Survival Versus Chemotherapy

Published Sep 14, 2026

Summary:
  • In updated late-stage data, patients on gotistobart lived a median 18.5 months versus 10 months for those on standard chemotherapy.
  • Earlier readouts showed a 54% reduction in risk of death versus chemo, at a point when median overall survival wasn't yet reached.
  • BioNTech's ADRs climbed up to 7.9% in Monday's pre-market trading after the update.

Trial results shaping a big cancer bet

BioNTech's latest readout centers on patients diagnosed with squamous NSCLC that has metastasized and has worsened despite prior immunotherapy and chemotherapy. The company said patients taking gotistobart had a median overall survival of 18.5 months, while patients on the standard chemotherapy regimen lived a median 10 months. In earlier results, the drug cut the risk of death by 54% versus chemotherapy at a time when the trial hadn't yet hit median overall survival. BioNTech added that the safety profile matched prior reports and remained manageable.

What experts and the market are saying

Medical oncologist Rama Balaraman, who serves as a principal investigator in the trial, said, "Current survival expectations with established therapies remain less than a year, and despite numerous development efforts, chemotherapy has remained the standard of care in this setting for more than a decade." Balaraman also noted that, should the survival advantage be verified in the upcoming portion of the late-stage trial, the therapy might offer a chemo-free alternative and "transform the standard of care" for this patient population.

"BioNTech's gotistobart survival benefit in second-line squamous non-small cell lung cancer underpins our $1.6 billion non-risk-adjusted peak-sales scenario and could establish a chemo-free option by 2028 for an underserved population. Bristol's rapid Opdivo uptake after its 2015 approval in the same setting supports the potential for fast adoption, while consensus peak sales of $877 million leave room for a rerating if our scenario is correct." - click here for the full note

Shares reacted, with BioNTech's American depositary receipts up as much as 7.9% ahead of the open on Monday.

Pipeline and leadership shifts

The results support BioNTech's push to launch multiple oncology products by 2030 as Covid-19 vaccine revenue fades. Last month, the company hit turbulence as it scrapped a clinical trial testing a personalized colorectal cancer vaccine, a program buoyed by the success of Moderna and Merck in the same class for melanoma.

BioNTech is working with US partner OncoC4 on gotistobart. The therapy aims to boost the immune system's ability to fight cancer by stripping away cells that let tumors dodge the body's defenses. Meanwhile, the company is in the middle of a leadership transition: co-founders Ugur Sahin and Özlem Türeci are preparing to leave by year end for a new venture, and Guido Oelkers is slated to take over as chief executive by February at the latest.

What this means for your portfolio

If confirmed in the next phase, this survival edge could set up a chemo-free option for a tough, underserved lung cancer group by 2028, which is why investors perked up. But BioNTech's story is also about execution: new leadership, a mixed pipeline with both wins and setbacks, and the long road of regulatory milestones. Watching how the data mature and how the company navigates the transition will tell you whether today's optimism sticks.

New developments remind investors that disciplined planning helps preserve and grow capital. Our CEO Jaspreet Singh is hosting a FREE live investor workshop, How to Profit From A Dollar That's Losing its Value, on September 29th. Sign up free to join him live.

Why RAM Prices Are Soaring - and Where the Money Is Moving
  • Memory chips - the RAM inside phones, laptops, fridges, and trucks - are in a shortage Tim Cook called a 100-year flood, and some memory prices have climbed about 90% in a single quarter.
  • Four forces hit at once: AI demand, a production shutdown in 2023, build times that push any fix to 2028 at the earliest, and a bombed helium plant in Qatar.
  • The last two supply shocks ended in aggressive Fed rate hikes and market drops of around 45% and 20%, and this time Washington is spending heavily to bring memory production home.

How Is the Economy Doing? Washington Says It's Fixed, but the Numbers Don't Agree
  • Treasury Secretary Scott Bessent says the economy is fixed because lower earners' incomes are now rising faster than top earners'.
  • The Atlanta Fed and Bank of America show different numbers, and Hilton, Marriott, and McDonald's can't agree on what they're seeing either.

US National Debt Hits $40 Trillion: Why the Economy Hasn't Collapsed Yet

Your 401k Is Fueling the AI Bubble
  • About $10 trillion of 401k money sits in a $77 trillion stock market, mostly through target date funds and S&P 500 funds. Roughly 30% of every S&P 500 dollar lands in five AI-heavy tech stocks.
  • Four bubble signals run hotter today than before the 2000 crash: top-ten concentration, tech's share of the index, the Buffett Indicator, and how much of the market index funds own.

What Is Wealth Preservation? How To Protect Your Money From Anything
  • Wealth preservation is an investing strategy built around keeping the money you've already made instead of chasing growth.
  • It leans on assets that hold steady when markets fall - gold, Treasury bonds, and companies that keep earning through wars, crashes, and pandemics.

Why Is Everything So Expensive? Why Prices May Never Come Back Down
  • Official inflation is 3.4% and prices are up 32% since 2020, but rent (41%), gas (47%), car insurance (64%) and ground beef (79%) all outran the 28% median wage.
  • The Federal Reserve targets 2% inflation on purpose. Rising prices push extra dollars to investors and shrink the real cost of a $40 trillion national debt.
  • Investors who simply owned the S&P 500 gained about 150% over the same six years, and the Fed's September 16 decision will show whether it protects the dollar or the economy first.
